Are you talking about Brian's guitar. . . . during Freddies intro?
If so, That song was recorded and usually played in drop D. It means the guitars lower E string has to be re tuned to D. Brian used to do this during Freddie's intro. You may have heard him strumming a few strings to be certain he was in tune.
FBG is also played in drop D another song which required re tuning during Freddies's intro. On the Q+ tours Brian has used a second RS guitar ( usually a Fryer guitar called George Burns, but sometimes a green Guyton guitar, which is already tuned to drop D
They (Brain and Roger, but mostly Roger) also do noises when Freddie says something they agree with, like live at the bowl "I'm positively knackered *Bang Bang* i can tell you"
Roger also tries to hit the drum at Freddies highest Eh Oh note.
There's also a moment in Rock Montreal where Freddie tells the audience they can take off their clothes, at which point John starts playing the Under Pressure bass line and Roger chimes in with the drums.
